Clomiphene

Mechanism

• Partial estrogen receptor agonist

• Acts specifically in hypothalamus which antagonizes normal negative feedback inhibition
• Results in an ↑ release of LH and FSH from pituitary
• stimulates ovulation

Indications

• Infertility
• PCOS 

Toxicity

• Ovarian enlargement
• Multiple birth pregnancy
• Visual disturbances 
• Hot flashes
